چکیده
It is demonstrated that positive supercoiling affiliated with nucleosome formation can act as the driving force to repair the G-quadruplex, cruciform as well as a stable non-B DNA structure caused by peptide nucleic acid.

NAP1-Assisted Nucleosome Assembly on DNA Measured in Real Time by Single-Molecule Magnetic Tweezers
While many proteins are involved in the assembly and (re)positioning of nucleosomes, the dynamics of protein-assisted nucleosome formation are not well understood. We study NAP1 (nucleosome assembly protein 1) assisted nucleosome formation at the single-molecule level using magnetic tweezers. This method allows to apply a well-defined stretching force and supercoiling density to a single DNA mo...
متن کاملThe supercoiling state of DNA determines the handedness of both H3 and CENP-A nucleosomes.
Nucleosomes form the unit structure of the genome in eukaryotes, thereby constituting a fundamental tenet of chromatin biology. In canonical nucleosomes, DNA wraps around the histone octamer in a left-handed toroidal ramp. Here, in single-molecule magnetic tweezers studies of chaperone-assisted nucleosome assembly, we show that the handedness of the DNA wrapping around the nucleosome core is in...

متن کاملRight-Handed Nucleosome: Myth or Reality?
In their recent paper in Cell, Furuyama and Henikoff (2009) report that nucleosomes in centromeres may be right-handed, that is, they wrap DNA in a right-handed manner and induce positive supercoils. This raises intriguing new questions, such as how centromeric histone variants may be assembled into right-handed particles, and why chromatin would retain negative supercoiling in chromosome arms ...
متن کاملHistone H3-variant Cse4-induced positive DNA supercoiling in the yeast plasmid has implications for a plasmid origin of a chromosome centromere.
The Saccharomyces cerevisiae 2-μm plasmid is a multicopy selfish genome that resides in the nucleus. The genetic organization of the plasmid is optimized for stable, high-copy propagation in host-cell populations. The plasmid's partitioning system poaches host factors, including the centromere-specific histone H3-variant Cse4 and the cohesin complex, enabling replicated plasmid copies to segreg...
